The P1B-type ATPases serve as heavy metal ATPases (HMAs) classified into monovalent and divalent ion-translocating groups; this study focuses on divalent cations transporting HMAs in Triticum aestivum L. (Poaceae). The in silico findings predicted that TaHMA1, TaHMA2, and TaHMA3 protein members interact with other proteins linked to metal homeostasis and oxidative stress regulation, suggesting broader roles in signaling and stress responses. Wheat seedlings were exposed to four concentrations of zinc (Zn; 100−400 µM), cadmium (Cd; 50−200 µM), and cobalt (Co; 50−200 µM) for 14 days in a hydroponic system. Analysis revealed distinct metal-specific responses: low Zn briefly stimulated growth, Co revealed biphasic trends with partial tolerance, while Cd caused severe and persistent toxicity. Gene expression analysis by qRT-PCR revealed strong early induction of TaHMA1 and TaHMA2 members that declined under prolonged stress, while TaHMA3 members were consistently upregulated, especially under Co and Cd, indicating a major role in vacuolar sequestration and detoxification.
