Ehretia acuminata (Pudila), a Boraginaceae fruit valued for antioxidants, is traditionally consumed by tribal communities. This study evaluates its antidiabetic, anti-inflammatory, antihemolytic, antimicrobial, and photoprotective activities using Soxhlet extraction with solvents of varying polarity: petroleum ether, chloroform, ethyl acetate, ethanol, and water. The activities were assessed spectrophotometrically, while antimicrobial activity was determined using the zone of inhibition method. Acarbose (AC), sodium diclofenac (SD), ascorbic acid (AS), benzophenone (BP), and erythromycin served as positive controls. With an IC50 range of 245.18 ± 0.27–250.6 ± 0.33, the chloroform extract demonstrated the strongest antidiabetic efficacy, whereas the ethyl acetate extract had the least impact, ranging from 2389.96–2380.24. With an IC50 of 285.22, the ethyl acetate extract showed the highest potential in anti-inflammatory tests, while the petroleum ether extract showed the lowest, at 870.28 ± 0.25. Additionally, the ethyl acetate extract exhibited the highest antimicrobial activity (zone of inhibition 10–12.5 mm against various food poisoning microbes), the highest antihemolytic activity (IC50 80.19 ± 0.08), and the highest photoprotective activity with an SPF of 15.16 ± 0.02at 500 μg/ml. These results indicate that the various extracts of E. acuminata fruit possess significant biological activities, likely attributable to their rich phytoconstituent content. They are promising natural sources for functional foods, pharmaceuticals, and skincare.
