Phytochemical evaluation and quantification of Mangiferin in Glysikot tablet; an Aayurvedic proprietary anti-diabetic formulation

Glysikot Tablet (GT) is an important Ayurvedic proprietary drug used to treat diabetes and associated conditions. It is prepared out of specific parts of five medicinal plants such as Curcuma longa L. Phyllanthus emblica L. Terminalia chebula Retz. Salacia chinensis L. and Tinospora cordifolia (Willd.) Hook.f. & Thomson. Mangiferin is an important anti-diabetic compound reported from Salacia chinensis, one of the ingredient plants of Glysikot tablet. In this study, quantification of Mangiferin has been done in Glysikot tablet using validated HPTLC technique in terms of linearity, precision, accuracy, limit of detection (LOD), robustness and limit of quantification (LOQ) in accordance with the International Council for Harmonization (ICH) Guidelines 2023. The result showed that Glysikot tablets contain 0.113% and 0.161% of Mangiferin in ethanol and hydro alcohol extracts respectively. Both ethanol and hydroalcohol extracts of Glysikot tablet showed a standard recovery of average 99% with a low percentage Relative Standard Deviation (RSD). Spectrophotometric estimation of major groups of phytochemicals showed that GT is rich in polyphenolics and alkaloids. In the present study, quality standardization of an important proprietary herbal medicine has been done with chromatographic and spectrophotometric techniques which will be beneficial for ensuring the quality and therapeutic value of this herbal formulation.
