Genetic diversity in plants refers to the genetic variation between the different species of plants. It is important to research the factors that can influence genetic diversity variance as well as the methods for determining genetic diversity. According to evolutionary changes, like genetic drift which involves the individuals mating randomly within the population, change in their allele frequencies, as well as the founder effect which results in the individuals of the species with new genetically information and a modified population emerges from the larger population. It also represents the number of genetic alterations in populations and species. The proper plant characterization and identification are crucial for using plants in the development of product, identifying their significant traits, and conserving plant species for the biodiversity preservation. In this chapter, the discussions are made on the different techniques of DNA sequencing followed by introduction of the PCR-based techniques. Among them, the mostly used molecular markers are SSR, RAPD, RFLP, AFLP, and ISSR. This chapter will be helpful for the proper knowledge and also to understand the applications of the molecular techniques.
