Oral administration of DSPE-PEG-cholic acid-modified ACDVs loaded with siLIMA1 for anti-MASH therapy

Metabolic dysfunction-associated steatohepatitis (MASH) is a chronic liver disease that lacks effective drug therapy. LIM domain and actin binding 1 (LIMA1) gene silencing acts as a promising therapy for MASH. The extruded red blood cell extracellular vesicles (artificial cell-derived vesicles, ACDVs) represent an ideal carrier for drug delivery. In this study, we successfully constructed distearoyl phosphatidylethanolamine (DSPE)-polyethylene glycol (PEG)-cholic acid (CA)-modified ACDVs loaded with LIMA1 siRNA (CA-ACDVs @siLIMA1). The nanoparticles were delivered to the liver via oral gavage, effectively silencing the expression of the LIMA1 protein in the liver and inhibiting the progression of MASH. Oral administration of CA-ACDVs demonstrates favorable safety, stability, and liver-targeting capabilities, making it a promising siRNA delivery method for the treatment of MASH.
