Autoantibodies against TRIM46 identified in a dog suffering from suspected meningoencephalomyelitis of unknown origin

Meningoencephalomyelitis of unknown origin (MUO) is a group of debilitating inflammatory central nervous system diseases with a reserved prognosis. Diagnosing MUO poses a challenge in clinical practice due to the lack of definitive biomarkers. We investigated a dog with MUO for the presence of autoantibodies targeting neuronal tissue in serum and cerebrospinal fluid, using indirect immunofluorescence assays. Autoantibodies targeting tripartite motif‐containing protein 46 (TRIM46), a brain‐restricted intracellular neuronal protein, were discovered using a tissue‐based assay and subsequently confirmed with a cell‐based assay. In humans, autoantibodies against TRIM46 are thought to be an expression of an aberrant immune response, leading to anti‐TRIM46‐encephalitis. Further, anti‐TRIM46‐encephalitis is often associated with underlying neoplasia. This is the first reported case of an intracellular neuronal target for autoantibodies discovered in a dog with MUO and suggests similarities to humans with autoimmune encephalitis subtypes and a possible field for research into specific clinical biomarkers.
