Rhodanine (derived from thiazolidine), a heterocyclic compound, plays an essential role in the biological system of humans.Its derivatives are present in drugs and are used in antibiotics, antiviruses, antidiabetics, and antifungals.We hypothesize that the shape-controlled synthesis of Poly-Rhodanine will provide an exciting perspective for diagnosing and treating diseases, including cancer.In our research, we used PolyRhodanine Copper NanoComposite which was synthesized in a single-step oxidation-reduction reaction in the presence of transition metals in the microwave in our chemistry laboratory.Subsequently, we tested our compound in a human lung cancer cell line, namely A549, to measure cancer cell viability by the colorimetric MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay.We also tested for the antimicrobial property of the newly synthesized compound against gram-negative E.coli bacteria by the commonly used Broth dilution method.For MTT assay, 20,000 A549 cells were plated in each well of a 96-well plate and treated with varying doses of the compound for 48 hours to investigate the viability of lung cancer cells.Our results showed viability of A549 cells decreased in a dose dependent manner with treatment concentrations from 0.01mM to 1mM in comparison to cells in the DMSO control treatment group.Interestingly, no such effect of the compound was found on HEK293 cells, which are known to be non-cancerous.With broth dilution method, we distinctly observed a dose dependent decrease in the number of viable E.coli cells depicting strong anti-microbial property of our newly synthesized compound.We could also identify the minimum inhibitory concentration for inhibiting growth of bacterial cells.Current studies are focused on gene expression studies, for understanding the specific gene target(s) of this compound.We would like to thank Qiaxian Johnson and Dr Bhanu P Chauhan from the Department of Chemistry, William Patterson University, Wayne, NJ, for the microscopic images.
