The eye and ear are complex special sense organs composed of highly specialised structures. The eye is the only special sense organ routinely evaluated in general nonclinical toxicity studies. Toxicity studies for the development of new ophthalmic or otic test articles and novel delivery techniques are crucial to improve options for the treatment of ocular and otic diseases. All components of the eye and ear may be susceptible to a wide variety of toxic changes following systemic or local exposure to test articles. Therefore, a good understanding of the normal comparative anatomy and histology of the eye and ear in laboratory species, along with an awareness of potential background/spontaneous findings and procedural/iatrogenic-related changes are critical when evaluating these special sense organs in the context of nonclinical toxicity studies. This chapter presents an overview of the normal anatomy, histology and physiology of the eye and ear, and provides comprehensive descriptions of various test articles-related toxic changes with specific examples in the most commonly used species in nonclinical toxicity studies. Examples of spontaneous/background findings and iatrogenic findings are also included.
