Développement et validation d’une méthode verte de dosage du paracétamol dans les formulations pharmaceutiques par chromatographie liquide haute performance

Abstract: High-performance liquid chromatography (HPLC) is the most widely used analytical technique for drug quality control. However, solvents commonly used are known hazardous for both analysts and environment. The aim of this work was to develop a green HPLC method for the determination of paracetamol in various pharmaceutical formulations. Ethanol, which is a green solvent, was used as organic solvent. A C18 column and a mobile phase consisting of 10% of ethanol and 90 % of acidified water (with acetic acid 10 mM), were employed. The flow rate and the wavelength were set at ,8 mL/min and 254 nm respectively. The paracetamol retention time was 7.56 minutes. The validation study showed that the developed method was specific, linear (R² greater than 0.99), fair (relative biases were less than 3%), precise (coefficients of variation for repeatability and intermediate precision were less than 3%) and accurate (tolerance intervals were within acceptance limits of 10 %) in the concentration range between 20 and 60 µg/mL. The greenness of the method was demonstrated using two different tools. The developed and validated green method was successfully applied to the analysis of different samples collected in the town of Bobo-Dioulasso in Burkina Faso.
