Design and Development of Nanoparticle Based Nanogel Formulation of Bixa Orellena Seed Extract

Background: Bixa orellana is known to contain Bixin, which has been known for its antifungal, antibacterial, and anti-inflammatory properties. Topical drug delivery is a widely utilized approach for achieving defined and sustained release of drugs at the intended site of action. Objective: The goal of the study is to prepare a nanoparticle based nanogel formulation from Bixa orellana seed extract. The present research work involves the synthesis of silver nanoparticles using an environmentally harmless and economically efficient method, utilizing the extract obtained from the seeds of Bixa orellana. Method: Silver nanoparticles (AgNPs) were prepared by green synthesis method using different concentrations of AgNO3 and concentration of extract at 500 rpm. Further nanogel was formulated by using hydroxypropyl methylcellulose (HPMC) K 15 and carbapol 940. Prepared formulation was evaluated for FTIR analysis, pH, viscosity, spreadability, and extrudability of gel. Result: From the results of FTIR Spectra it was observed that there is no any chemical interaction between drug and excipients. The pH, viscosity, spreadability, and extrudability of gel were found to be 6.45±0.50, 6515±1000 cps at 200 rpm, 5.3±0.12 cm, 0.5 cm respectively. The results of the in vitro drug release investigation reveal a significant drug release of 82.54% within a 10 h period with 1 gm of HPMC K 15, while Carbapol exhibited a moderate level of drug release. Conclusion: From in vitro drug release it found that HPMC K 15 and carbapol 940 exhibit controlled drug release from nanogel formulation while HPMC K15 at 1 gm concentration showing significant drug release. In future perspective the in vivo investigation of nanogel formulation may be a fruitful discovery against various activities of Bixa orellana with respect to topical delivery.
