Role of Non-Flavonoidal Agents in Alzheimer's Disease

Among older individuals, Alzheimer’s disease is the most prevalent cause of dementia, accounting for approximately 60% to 70% of all cases. As a neurodegenerative disorder, Alzheimer’s has become a focal point in neuroscience research, aiming to understand the cellular changes and underlying mechanisms involved. Natural compounds, such as flavonoids, are believed to have the ability to cross the blood-brain barrier and have shown potential in the central nervous system. Consequently, these chemical components are being investigated for their effectiveness in slowing down the progression of neurodegenerative diseases. Diagnosis of Alzheimer’s disease in dementia patients is based on the presence of neurofibrillary tangles and senile plaques, which are characteristic features of the condition. Although there are drugs like donepezil currently used to treat these ailments, they often come with adverse effects, leading healthcare professionals to explore alternative therapeutic options. While there is currently no definitive cure for Alzheimer’s disease, pharmacological interventions are available to slow down its progression. Traditional herbal medicines used in many countries have been 190 employed to prevent or treat neurodegenerative disorders and some have been formulated as nutraceuticals or dietary supplements. Non-flavonoid polyphenolic compounds are a specific category of natural compounds that have shown promise in the treatment of Alzheimer’s disease. These compounds and their derivatives act as antioxidants, increase the expression of anti-inflammatory mediators, reduce A β aggregation and deposition of tau proteins, thereby protecting neurons and improving the condition of Alzheimer’s disease. This chapter provides a brief overview of the pathology of Alzheimer’s disease and explores the potential of non-flavonoid polyphenols and their derivatives as active agents against the disease.
