Understanding mechanisms of non-responsiveness to antidepressants is critical to discovery of new, effective treatments for treatment-resistant depression, a disease that affects more than one-third of patients with depression. This study assumes that in depressive state the activity of ventral hippocampus-medial prefrontal cortex (vHPC-mPFC) pathway is attenuated, and the resistance to antidepressant treatment may arise from the inability to activate this pathway. We used ibotenic acid (IBO, 5 µg/0.5µl, bilateral) lesion in vHPC in Wistar and Wistar-Kyoto (WKY) rats to investigate an involvement of vHPC-mPFC pathway in the action of venlafaxine (10 mg/kg, IP), ketamine (10 mg/kg, IP) and Deep Brain Stimulation (DBS) in left mPFC in animals exposed to the chronic mild stress (CMS) model of depression.
