Supplemental Figure 1. (A) Annexin V/7AAD viability staining of ATC cells (U-Hth 104 cell line), non-malignant thyroid cells(non-toxic goiter), immortalized fibroblast cells (BJ cell line) autologous B cells and T cells cultured with or without expanded NK cells at a 1:1 ratio for 4 h. (B) Mean fluorescence intensity (MFI) of NKG2D-ligands on viable tumor cells from ATC FNA. MFI values from NKG2D-ligand staining have been subtracted with isotype control MFI values (C) Specific lysis of the ATC cell line LUTC-17 by autologous NK cells in a 5h cytotoxicity assay. Blocking antibodies targeting ULBP2 or NKG2D was added 30 minutes prior and during the cytotoxicity assay. Effector-to-target ratio = 2:1. The experiment was performed once.
