The opioid system modulates numerous physiological functions with mu opioid receptors mediating opiate analgesic and euphoric properties. Functional crosstalk between mu and delta opioid receptors was postulated through heteromerization of the two receptors. Signaling and trafficking properties of mu-delta heteromers have been extensively studied in transfected cells but little is known about their functional impact in native tissue or about their distribution in the nervous system. Using double knock-in mice co-expressing functional fluorescent mu and delta receptors, we mapped mu-delta neuronal co-localization in the brain in basal and pathological conditions. We review here the technological improvements developed to optimally visualize the two receptors in vivo.
