Introduction and Aims: To explore the effect and mechanism of fluvastatin on the expression of fibronectin(FN) in human peritoneal mesothelial cells(HPMC) induced by high-glucose peritoneal dialysate. Methods: Cultured HPMC were randomly divided into si &hebrewnun;groups of control(a 1:1 mixture of DMEM and total culture fluid), high-glucose peritoneal dialysate(a 1:1 mixture of 4.25% peritoneal dialysis solusions and total culture fluid, HGPDS), HGPDS plus fluvastatin 10-8mol/L, 10- 7mol/L, 10-6mol/L, HGPDS plus GSK650394 10-5mol/L(the competitive inhibitor of SGK1), fluvastatin 10-6mol/L and GSK650394 10-5mol/L . The cellular viability were detected by MTT colorimetry, the mRNA expressions of serum and glucocorticoid -inducible kinase 1(SGK1) and FN were detected by RT-PCR. Results: The results of MTT revealed that the groups of DMSO and different concentrations of fluvastatin had no significant effect on viability of HPMC(P > 0.05), while lower HPMC viability was found in the groups of HGPDS, HGPDS plus different concentrations of fluvastatin and HGPDS plus GSK650394 (P < 0.05). Compared with the HGPDS group, the viability of HPMC was improved significantly in HGPDS + fluvastatin 10-6mol/L group and HGPDS + GSK650394 group (P < 0.05)at the time of 24h.Compared with the normal control group, the mRNA expression of SGK1 and FN increased significantly in HPMC treated with HGPDS, which peaked at 1h and 6h respectively. GSK650394 significantly decreased the high mRNA expression of SGK1 and FN induced by HGPDS (P < 0.05), and the fluvastatin significantly inhibited the activations of SGK1 and FN mRNA induced by HGPDS in a dose- dependent manner(P < 0.05). Conclusions: High-glucose peritoneal dialysate can increase FN expression in human peritoneal mesothelial cells, which can be decreased by fluvastatin. The role of fluvastatin may partially achieved through the signal pathway of SGK1.
