To clarify the chronic morphine-induced genes, I tried to construct the subtractive eDNA library from the mice amygdala complex treated with saline and chronic morphine.Morphine hydrochloride was injected to male ddY mice (6week-old) subcu-taneously twice a day for 7 days as describe d (1).mRNAs were extracted from amygdala complex at 16 hours after the last administration.By using differential hybridization method, I finally isolated 11 positive eDNA clones from 1.8 x 10 4 plaques of the subtractive e DNA library.In situ hybridization analysis revealed that the increase of 01 mRNA was detected at the ~ basolateral amygdala nucleus only by chronic morphine treatment.It was completely blocked by co-administratio n of naloxone Furthermore, 01 mRNA was localized in the brain regions such as cerebral cortex, dentate gyrus, hippocampus (CA1-3), cerebellum ( Bergman glia) and locus coelurus.Northern blot and RT-PCR analysis suggested that 01 mRNA might be localized in glial cells.These results indicate that chronic morphine administration elicits a change of gene expressions in glial cells of amygdala complex and that glial cells have an important role for inducing tolerance and dependence ..
