Purpose: Platelets play a vital role in hemostasis and thrombosis. Catecholamines have a profound effect and alpha2B–adrenergic receptors (α2B–ARs) are implicated in the pathophysiology of platelet aggregation. In this report, we investigated the effect of systolic blood pressure (SBP) levels on α2B–ARs gene expression in platelets from patients with essential hypertension compared to normal individuals. Methods: Thirty six uncontrolled (office SBP >140 mmHg and ambulatory daytime SBP >135 mmHg, 22 males, aged 55±9 years) and 32 well controlled (office SBP <140 mmHg and ambulatory daytime SBP <135 mmHg, 21 males, aged 58±8 years) essential hypertensive patients were included in the study. Twenty four healthy individuals (12 males, aged 53±10 years) were served as control group. Blood samples were obtained from which platelets were isolated. The α2B-ARs gene expression in platelets was examined by real time PCR. Results: Patients with uncontrolled essential hypertension showed a significantly increased expression of the receptor in the platelets (38.6±34.6 in normal, 37.2±24.6 in controlled hypertension versus 77.4±59.7 in uncontrolled hypertension, p<0.01 for both). Notably, patients with adequately controlled hypertension did not show any significant difference from healthy individuals. The α2B-ARs gene expression in platelets was significantly correlated with SBP levels (r=0.51, p<0.001).
