The Use of Vacuum in Processing Intact Ticks for Histology

There are many properties inherent to the nature ofixodid ticks which create obstacles in preparing them for fine sectioning. The impermeability of the tick cuticle prevents fixation when ticks are simply submerged in the fixatives for various lengths of time. This situation leads to the use of dissected tick tissues for the study of different aspects of tick anatomy and histology but also the biology of the agents for which ticks are vectors. Although preparation ofhistological sections of dissected tick tissue has been useful, the anatomical relationship is lost and normal cavities between organs, filled, in many instances, with hemolymph as well as connective tissue, are disturbed or missing. Larvae and engorged females of a Puebla strain of Boophilus microplus (Canestrini) ticks were used in our study of vacuum-assisted fixation.
